The influence of dietary casein level on tumor induction with 2-acetylaminofluorene.

چکیده
INTRODUCTION The present experiments were conducted to de termine the carcinogenicity of 2-acetylaminofluorene when it was fed in partially purified diets that varied in casein level between 9 and 60 per cent. Studies involving this broad range of dietary protein level have not been reported previously with this carcinogen. Harris (7) concluded that variations in dietary protein level had no appreciable effect on the pro duction of liver tumors with 2-acetylaminofluorene in rats. Purified diets containing 13 and 20 per cent of casein were employed. Morris and associates (10) found that rats developed more tumors when purified diets containing 18-24 per cent of casein were used than when the diet contained 12 per cent of casein. In these studies, it was noted that dietary pro tein levels in the range used by the above investi gators had no influence on tumor induction by 2-acetylaminofluorene. However, it was observed that diets containing 40 or 60 per cent of casein had a definite protective effect against the induc tion of mammary tumors and possibly other types of tumors.

INTRODUCTION Investigations in this laboratory on the carcino genic action of 2-acetylaminofluorene have re vealed that the type of diet fed with the carcinogen can influence the incidence and types of tumors in duced in rats. Female rats of the Alabama Experi ment Station (AES) strain uniformly developed mammary tumors in about 4 months when they were fed this carcinogen on a semi-purified die...

In 1948 it was reported from this laboratory (4) that the feeding of @-acetylaminofluorene to wean ling female rats on a semi-synthetic diet produced a 100 per cent incidence of mammary tumors†" a much higher incidence than had been reported by other investigators. Following these observations, studies were initiated to determine whether or not modification of the diet would influence the tum...

In the first study of the toxicity and carcino genic activity of 2-acetylaminofluorene (AAF), Wilson, DeEds, and Cox noted that the compound caused a profound inhibition of growth, when it was fed in a 10 per cent casein diet to albino rats (21). Inhibition of the growth of the rat has also been obtained with a number of carcinogenic aro matic hydrocarbons and with p-dimethylaminoazobenzene (11...

We explored, in rats, the conditions underlying the induction of liver tumors by ./V-hydroxy-./V-2-fluorenylacetamide (N-OH-FAA) in terms of the participation of an ultimate carcinogen in the form of the sulfate ester. Male rats were fed 2 dose levels, i.e., 0.0213%(0.89 mmole/kg) and 0.032%(1.34 mmoles/kg) of N-OH-FAA in the diet, with or without 0.89% (59 mmoles/kg) p-hydroxyacetanilide, and ...
